A polypeptide which is a mutated mannanase, whose sequence is derived from a native mannanase of the filamentous ascomycete coprophile fungus Podospora anserina, and being characterised by an increased catalyst efficiency of at least 25%. Also, a polynucleotide encoding the polypeptide, a vector including the polynucleotide, a host cell including the vector or the polynucleotide, and a composition including the polypeptide, the polynucleotide, the vector or the host cell. Methods of using the polypeptide, the polynucleotide, the vector, the host cell or the composition of the invention for degradation of compounds including mannans, in particular lignocellulosic biomass, as well as various possible uses of the polypeptide, the polynucleotide, the vector, the host cell or the composition.
