Disclosed are a system and methods for identifying JAK2-specific polynucleotide sequences in a biological sample. Also disclosed are oligonucleotide primer and probe comopositions for detecting mutations in a JAK2 polynucleotides, and the JAK2.sup.V617F mutation in specific, as well as systems, diagnostic kits and articles of manufacture comprising JAK2-specific primer and probe compositions.
